Automated Gentle Media Exchange for Loosely Adherent HEK-293T Cells with the Reagent Exchanger (RX)

Abstract

Due to its inherent gene expression profile, HEK-293T is a commonly used cell line known for its tendency to detach easily from the growth surface. This characteristic makes careful handling essential during in vitro 293T maintenance, especially to avoid undesired cell loss during media exchange. While manual handling can sometimes cause unintended detachment, automated systems can provide consistent and gentle liquid handling through stable flow rate control. In this report, we demonstrate that the Formulatrix® Reagent Exchanger (RX) can reliably perform gentle media aspiration and dispensing, with an average cell detachment of only 5% cell loss from the initial 293T culture confluency, compared to 10% average cell loss with manual hand pipetting. Moreover, the RX offers multiple nozzle options, allowing both safe and efficient maintenance of 293T cultures in many SBS-format plates.
