Extracellular vesicles (EVs) hold significant promise in diagnostics, therapeutics, and drug delivery. However, their isolation and formulation pose significant challenges due to the critical need for high purity, particle integrity, and sample recovery. This study demonstrates the effectiveness of a combined approach utilizing IZON's qEV columns for EVs purification and Formulatrix's μPulse - TFF system for subsequent formulation. EVs were isolated from human plasma by qEV 10 columns and characterized by tunable resistive pulse sensing (TRPS). Following purification, the vesicles were concentrated using the μPulse. Post-concentration TRPS analysis revealed minimal changes in EVs size, with only a 2.36% and 3.35% change in mean and mode particle diameter, respectively, indicating the gentleness of the μPulse.
The μPulse system also demonstrated high particle retention (97-98%) during the concentration process. This integrated approach provides a robust and efficient workflow for EVs isolation and concentration, paving the way for advancements in EV-based diagnostics and therapeutics.
