Turn Unpromising Conditions into Hits Using Iterative Screen Optimization (ISO)
Rock Maker simplifies Iterative Screen Optimization (ISO), an automated optimization process that helps researchers generate crystals from conditions that may not initially look promising. ISO works by systematically adjusting the precipitant concentration in follow-up experiments to steer conditions into the nucleation zone of the protein solubility curve.
Using a premade screen, you begin with an initial experiment and score each drop as Clear, Crystal, Light Precipitate, or Heavy Precipitate. Based on these scores, Rock Maker automatically creates a new experiment by adjusting the precipitant concentration for each condition. In each subsequent ISO round, Rock Maker uses results from the last two iterations to fine-tune the next set of concentrations.
